(uint16_t*) is the cast. You are "transforming" the number 0x550000 in a memory address (pointer)... But to access the content of the that address you need to use *... Change a bit the code, may be it's more compreensive: #define memaddress (uint16_t*)0x550000 // ... *memaddress = 33;
